چکیده

To calculate the RDF, for each time frame we used the centers of the neurofilaments in the domain [−0.2μm, 0.2μm] × [−0.2μm, 0.2μm] as reference points. We chose to use reference points in a smaller domain in order to avoid boundary effects. For each reference point we binned the center-to-center distances (r) between the reference neurofilament and all other neurofilaments with a bin size ∆r = 1nm, and normalized by the factor 2πr∆rρN where ρN is the density of neurofilaments in the whole domain. We then took the average over all reference points for each time frame and then over all time frames to obtain g(r). We note that g(r) is noisy due to under sampling and the small bin size (1 nm). Averaging over more time frames or using a larger bin size gave smoother g(r). To compute the OPD pn, we used circular windows with fixed radii 60 nm as in [1]. For each time frame, we sampled centers of 10nN circles according to the uniform distribution in the domain [−0.3μm, 0.3μm] × [−0.3μm, 0.3μm] and calculated the particle occupancy number for each circle. We then produced a histogram of occupancy numbers obtained for all circles and normalized it by the total number of circles to obtain pn.

A Stochastic Multiscale Model That Explains the Segregation of Axonal Microtubules and Neurofilaments in Neurological Diseases

The organization of the axonal cytoskeleton is a key determinant of the normal function of an axon, which is a long thin projection of a neuron. Under normal conditions two axonal cytoskeletal polymers, microtubules and neurofilaments, align longitudinally in axons and are interspersed in axonal cross-sections. However, in many neurotoxic and neurodegenerative disorders, microtubules and neurof...

Segregation Produced by P,p’-iminodipropionitrile: Evidence for the Association of Fast Axonal Transport with Microtubules

The subperineurial injection of /3#‘-iminodipropionitrile (IDPN) into rat sciatic nerves resulted in focal disorganization of the axonal cytoskeleton characterized by segregation of neurofilaments and microtubules. Shortly after injection, microtubules clustered together to form a central channel, while neurofilaments became chaotically arrayed between the microtubule channel and axolemma. Elec...

Neurofilaments bind tubulin and modulate its polymerization.

Neurofilaments assemble from three intermediate-filament proteins, contribute to the radial growth of axons, and are exceptionally stable. Microtubules are dynamic structures that assemble from tubulin dimers to support intracellular transport of molecules and organelles. We show here that neurofilaments, and other intermediate-filament proteins, contain motifs in their N-terminal domains that ...

B cell Therapy to Treat an Axonal Neuropathy in Mixed Connective Tissue Disease

The B cell is a vital contributor to humoral immunity. The B cell-specific antigen CD20 is expressed during B cell development, starting at the pre-B cell level and persists through B cell differentiation, but is lost during terminal differentiation to plasma cells. Rituximab is a monoclonal antibody that destroys both normal and malignant B cells that have CD20 on their surfaces and is ther...

Redistribution of proteins of fast axonal transport following administration of beta,beta'-iminodipropionitrile: a quantitative autoradiographic study

Beta,beta'-iminodipropionitrile (IDPN) produces a rearrangement of axoplasmic organelles with displacement of microtubules, smooth endoplasmic reticulum, and mitochondria toward the center and of neurofilaments toward the periphery of the axon, whereas the rate of the fast component of axonal transport is unchanged. Separation of microtubules and neurofilaments makes the IDPN axons an excellent...
